Label-free Quantification

Label-free quantification is a mass spectrometry analysis of peptides through liquid-mass spectrometry technology. The quantification of unlabeled peptides can be performed by extracting the peptide signal intensity or by spectral counting. The ion signal intensity approach uses the extracted chromatographic area to compare peptide abundances across samples. Spectral counting totals the number of times a peptide is selected for fragmentation and identified in a data-dependent acquisition experiment. This method does not need to use expensive stable isotope labels as internal standards, requires a small amount of samples, and has low cost.

Isobaric Labeling

Isobaric labeling is a mass spectrometry strategy used in quantitation. Peptides are labeled with various chemical groups that are (at least nominally) identical masses (isobaric), but vary in terms of distribution of heavy isotopes around their structure. These tags, commonly referred to as tandem mass tags, are designed so that the mass tag is cleaved at a specific linker region upon high-energy CID (HCD) during tandem mass spectrometry yielding reporter ions of different masses. Isobaric labeling has been successfully used for many biological applications.

Isotope dilution mass spectrometry avoids the influence of matrix effects and analysis conditions, has special advantages in quantitative analysis and accuracy, and has been widely used in the field of quantitative analysis of biomolecules. Stable isotope-labeled peptides are widely used as important internal standard reagents in dairy product detection, biological protein quantification, protein sequencing, drug metabolism, and food allergen detection. This method has the advantages of high throughput, good repeatability, accurate quantification, reliable identification results, and simple operation.
