Venom N-Glycan Quantitative Analysis

The complexity of the venom is due to post-translational modifications (PTM), including proteolytic processing, oligomerization, and glycosylation. N-glycosylation is the main PTM of the venom, which adds another level of complexity to the venom's proteome and may contribute to the unique biological activity of the venom. Therefore, it is necessary to study the N-glycan of venom. In addition to the qualitative analysis of venom N-glycan, Creative Proteomics uses isotope labeling technology to provide relative and absolute quantification services of venom N-glycan based on MS.

Relative Quantitative Analysis of Venom N-Glycan

MS-based relative quantitative N-glycan is widely used in venom glycomics research, generally using isotopic labeling or isobaric tag. We are confident to provide professional and reliable quantification services of venom N-glycan, our analytical service is dedicated to improving the quantification accuracy, sensitivity and simplicity of N-glycan in venom.

  • Isotopic labeling
This is a method that uses stable isotope labeling of N-glycan in the venom for relative quantification, which can realize the simultaneous quantification of acid sugar chains and neutral sugar chains It can also be used to qualitatively glycosylate the venom and determine the binding sites of glycosyl groups on proteins and lipids.
  • Isobaric tag
It is an isobaric labeling method that can determine the number of N-glycan from different sources in one experiment by mass spectrometry. A mixture of N-glycan is obtained by hydrolyzing protein samples, all samples are labeled with different reagents and mixed, and the N-glycans are analyzed by tandem mass spectrometry to obtain mass spectra.

Absolute Quantitative Analysis of Venom N-Glycan

On the basis of relative quantification, we can provide possible absolute quantitative analysis when chemically pure standard N-glycan are available. We determine the true ratio of different N-glycan structures in the venom for our customers, and are committed to improving the quantitative accuracy.
