Mevalonic acid (MVA) is an important organic compound in biochemistry. The name is a contraction of dihydroxymethylvalerolactone. The carboxylate anion of mevalonic acid, which is the predominant form in biological environments is called mevalonate, and is of major pharmaceutical importance. Drugs, such as the statins which is biologically active to lower levels of cholesterol, stop the production of mevalonate via inhibiting HMG-CoA reductase.

MVA is a key step in cholesterol biosynthesis; it is an important intermediate product of cholesterol and terpenoid synthesis, and plays an important role in regulating cell proliferation and collagen synthesis. MVA stimulates HMGR expression and plays a key role in regulating cell growth, but studies have shown that high cholesterol levels inhibit cell growth via HMGR feedback inhibition in vitro. Intramuscular lipid contained more cholesterol than either subcutaneous fat layer.
Actually, MVA is a precursor in the biosynthetic pathway referred to as the mevalonate pathway that generates steroids and terpenes. MVA is the primary precursor of IPP, which is in turn the basis for all terpenoids. Mevalonic acid is chiral and the (3R)-enantiomer is the only one that has biologically activity.
The mevalonate pathway, also referred to as the HMG-CoA reductase pathway or isoprenoid pathway, is a very important metabolic pathway present in archaea, eukaryotes, and some bacteria. This pathway generates two five-carbon compounds: dimethylallyl pyrophosphate (DMAPP) and isopentenyl pyrophosphate, which are used to produce isoprenoids, a wide-ranging class of over 30,000 biomolecules such as cholesterol, vitamin K, coenzyme Q10, heme, and all steroid hormones. The mevalonate pathway starts with acetyl-CoA and ends with the generation of IPP and DMAPP. It is most famous as the target of statins, a class of cholesterol lowering drugs. Statins inhibit HMG-CoA reductase within the mevalonate pathway.
