As the major component of membranes and energy resources, cellular lipids have an important role for both cellular and physiological energy homeostasis. The main biological functions of lipids include storing energy, signaling, and serving as structural components of cell membranes. The composition of lipids varies among different tissues. In animal tissues, the structural lipid components, such as the phospholipids, are rather constant in composition under the normal physiological conditions. In contrast, the proportions of the simple lipids, especially the triacylglycerols, can vary significantly according to the physiological condition and nutrition of the animal. The compositions of the plasma lipids are of particular importance because they feed fatty acids to all tissues. Brain phospholipids have a higher proportion of phosphatidylserine than other tissues. In liver, PC and PE are very abundant.
Since lipid metabolism is regulated within a network, changes of any synthesis or degradation pathway might result in the change of the entire lipid homeostasis of the network. Untargeted lipidomics analysis provides deep insights into the mechanisms of the entire system after particular stimulus. Furthermore, under certain disease or pathogenic conditions, determining the altered lipid species and quantity will potentially reveal the mechanisms of the disease. Lipidomics can serve as a powerful tool for screening and validating hypothesis. Moreover, lipidomics can be very useful in biomarker discovery in different diseases, including diabetes and obesity, cardiovascular diseases, nonalcohol fatty liver disease, psychosis, and cancers.
