Introduction

Protein glycosylation is a functionally important but very complex type of post-translational modification. Therefore, comprehensive characterization of glycan structures and their site-specific locations on proteins at the proteomic scale remains an experimental challenge. Glycosylation, an essential post-translational modification of proteins, plays a role in a variety of biological processes such as controlling protein folding, regulating immune responses, and mediating interactions between host cells and enzymes, antibodies, receptors, and various pathogens. Glycosylation, which is typically low in abundance in biological systems, exhibits dynamic changes not only in the size and composition of glycan chains, but also in sialylated linkages. Mass spectrometry is a high-throughput method and provides a useful tool to decipher these dynamically changing structural details to understand the function of protein glycosylation.
