Antibody CDR Sequencing

Antibody CDR Sequencing

Service Details

Introduction

CDRs, a variable amino acid sequence, stand for complementarity determining regions that folds into loops capable of binding to antigenic amino acid sequences. The complementarity determining regions (CDRs) of antibodies are highly variable, and the hypervariable regions in the variable regions of the heavy and light chains of the antibody (Ab) constitute the antigen (Ag) binding point of the antibody molecule, because the antigen binding point is closely related to the antigen surface. The structure of the antibody is complementary to each other, so the hypervariable region is also known as the complementarity-determining region (CDR) of the antibody molecule. Comparing the amino acid sequences of different antibody V regions, it is found that VH and VL each have 3 regions of amino acid composition and sequence. Particularly variable, these regions are called HVRs. The three HVRs of VH and VL together constitute the antigen-binding site, which is also called the CDR.

Our Services

Creative Proteomics has established the mass spectrometry-based antibody sequencing platform by adopting DASS (Database Assisted Shotgun Sequencing) technology, combining the advantages of existing technologies to sequence antibody CDR regions.

Essentially, CDRs are new protein sequences. Routine identification of proteins is often performed by genomics and proteomic approaches that require database searches-more advanced techniques are required because antibody sequences are rarely found in databases.

The mass spec sequencing method established by Creative Proteomics uses multiple digestions to generate many overlapping peptides during fragmentation, allowing us to sequence very long unknown amino acid sequences. In addition, our experienced scientists combine high-quality MS/MS spectra with intelligent data mining technology, which makes CDR sequencing easy. Based on the power of this technology, we were able to sequence 20 kDa proteins with no homologues in the database.

Advantages of Our Services

  • For antibody complementarity determining regions (CDR regions), generate peptide sequences containing a single site 10 times, and pass high coverage to ensure accurate subsequent antibody sequence splicing.
  • The time spent on the DASS platform is less than half of that of traditional sequencing methods, and the full sequence determination can be completed within 3-10 working days after the sample is received.
  • low sample size.
  • Our DASS method is compatible with high throughput enabling accurate interpretation of sequences.
  • Scientists in charge of each project, with dedicated teams that perform quality control at every step of the process, and expert R&D teams dedicated to optimizing protocols to deliver higher quality data.
  • Custom services, including but not limited to antibody de novo sequencing and analysis, and in vitro optimization to meet your needs.

At Creative Proteomics, we provide professional antibody CDR sequencing services for global customers. Our scientists can also provide and help design the best solutions according to your specific requirements, and describe them in detailed project reports. Please feel free to contact us if you are interested in our services.
